The Human Technopole (HT) is a new interdisciplinary research institute in Milan, Italy, created and supported by the Italian government whose aim is to develop innovative strategies to promote human health through a multidisciplinary and integrated approach, combining genomics, computational and structural biology, neuroscience, and data and decisions sciences.

HT has the mission of conducting cutting-edge research but also to translate innovation into methods and tools available to the entire National scientific community.

To this end, HT is opening National Facilities that will provide state-of-the-art research infrastructure for Structural Biology, Light Imaging, Genomics, Genome Engineering, and Data Handling and Analysis.


The primary objective of the NF for Genome Engineering is to establish a disease modeling platform enabling the provision of a wide range of services in stem cell biology, differentiation and genome engineering.

The Facility is organized into four distinct units, each serving a specific purpose:
(1) Pluripotent Stem Cells (PSCs) Engineering Unit; (2) Cell Lines and Tissues Engineering Unit; (3) Model Validation; (4) Technology development.


The Human Technopole Foundation is recruiting a Manager to oversee the operations of two key infrastructure units within the National Facility for Genome Engineering.

This individual will be responsible for providing strategic guidance and efficient management for Unit One,
focused on the engineering of PSCs, and Unit Two, focused on the Engineering of immortalized/cancer cell lines and tissues.

Through the integration of advanced robotics, bioengineering technologies and a highly interdisciplinary approach, the Facility aims to deliver comprehensive services and facilitate the execution of complex, interdisciplinary genome engineering workflows.


Key tasks and responsibilities:


  • Establishing and managing the core functions of the PSCs and cell line editing units, integrating user requirements and translating them into high quality operations and services;
  • Coordinating and supervising junior staff;
  • Establishing and standardizing currently available gene editing technologies (CRIPSRcas, base and prime editors,) in pluripotent stem cells and cell lines;
  • Testing and benchmarking new technologies for the automation of the gene editing pipelines;
  • Overseeing and assisting with experiment validation and problem solving;
  • Designing and running gene editing experiments in synergy with other members of the Facility;
  • Developing innovative strategies in genome engineering;
  • Keeping detailed records of engineered cell line production and inventory;
  • Supporting the Head of the Facility in setting up the essential resources, participating in key aspects of general facility management and organization (e.g., managing inventories and stocks of samples, reagents, equipment, and materials).

Mandatory Requirements:

Ph.
D. in biological or life science fields;

  • At least 5 years of relevant work experience;
  • Track record in the following areas:
  • Experience with the complete portfolio of CRISPR-based genome editing techniques: from Knockout lines generation to engineering of specific
mutations and/or reporters, as well as gene expression manipulation throughCRISPRi and CRISPRa, and CRISPR screening approaches. o Mammalian cell culture techniques.

  • Familiarity with basic molecular biology and biochemistry methods (DNA and RNA manipulation, cloning, RT-PCR, immunoblotting and immunoprecipitation techniques);
  • Good knowledge of English.

Preferential requirements
-
Basic knowledge with prime-editing and base-editing methods;

  • Experience with embryonic and/or induced pluripotent stem cells;
  • Experience and skills in performing repetitive and specialized laboratory experimental procedures;
  • Previous experience in training and coordinating students and junior scientists.
