    Within the Computational Biology Research Centre, HT hosts a core experimental unit (DGEV) that provides support for the creation of novel molecular datasets from clinically annotated patient cohorts and is one of HT's leading resources for the generation of patient-derived model systems from human samples.

    About the position For the DGEV core unit, we are seeking a Technician/ Senior Technician in Cell and Molecular Biology to enrich the team by leveraging their expertise in drug screening.

    The Technician will work on culturing and manipulating patient-derived organoids to support the experiments of various research projects at HT, including flagship projects.

    The work includes setting up and maintaining experimental pipelines for drug screens with a focus on gastrointestinal organoids as well as human cell lines.

    As we look toward the unit's growth, we aim to expand the role to include Gene Editing and CRISPR Screening capabilities.

    The selected candidate will receive comprehensive training in these cutting-edge techniques to foster future developments within the unit.

    The Technician will collaborate with other members of different research groups, including other experimental researchers to generate large datasets, as well as computational scientists for the analysis of the data.

    They will work closely with a diverse team to formulate and validate hypotheses, offering an unparalleled opportunity for measuring and understanding disease dynamics.

    The DGEV unit benefits from accessibility to the HT National Facility for Genomics, providing state-of-the-art and innovative services ranging from high-throughput sequencing to multi-omics technologies.

    The candidate will also collaborate closely with the HT National Facility for Genome Engineering and Disease Modelling in the future.

    The successful candidate will report to Prof. Andrea Sottoriva, who is the Head of the Computational Biology Research Centre.

    Key tasks and responsibilities Main areas: Designing and performing drug screening experiments using human cancer model systems (patient-derived organoid cultures and human cell lines).Applying, generating, and optimizing protocols for human cell cultures.

    We aim to stay at the forefront of science in the field. Setting up single-cell perturbation screens.

    Performing meticulous sample tracking, storing and QC.Generating reliable data for publications in the assigned projects and recording all data and results in specified forms.

    Collaborating with the HT National Facility for Genomics for sample profiling.
    Working closely with colleagues from the HT National Facility for Genome Editing.

    Providing technical support and supervision to other laboratory members, including introduction, and teaching of new laboratory techniques and supporting of the use of lab equipment.

    Learning new techniques from other laboratory members.

    Liaising with the Centre's Lab Manager and other staff to write risk assessments and ensure that they are regularly updated.


    Additional areas:
    Designing and performing genome editing experiments using human cancer model systems.
    Applying, generating, and optimizing protocols for genome editing using CRISPR-Cas9 technologies.
    Setting up targeted and genome-wide CRISPR screens.

    Essential requirements A Bachelor degree in molecular biology, genetics, biochemistry, or in a related field of the life sciences is required.

    Considerable relevant laboratory experience in cell and molecular biology.
    Experience in mammalian cell culture and drug screening. Fluency in English – HT is an international research institute.
    Preferred requirements Documented hands-on experience with the application of cutting-edge methods in cell biology, including human organoids culture.
    Strong background in molecular biology, experience with viral and non-viral vector development, and clone screening.
    Experience in bacterial transformation, and restriction enzyme analysis.
    Familiarity with 10x single-cell genomics assays.


    Advantageous:
    Knowledge on flow cytometry technologies and applications.


    Advantageous:
    Experience in genomic techniques, transfection, and knockdown experiments (e.g., CRISPR/Cas9 genome editing, siRNA, shRNA, antisense and similar technologies).


    Advantageous:
    Knowledge or experience in genome-wide CRISPR-Cas9 screening with patient-derived model systems.


    Advantageous:
    Experience with handling large-scale projects is considered advantageous.
    Organizational and social skills Ability to manage competing priorities in a fast-paced environment and to work independently.
    Excellent organization and communication skills.

    Strong interpersonal skills demonstrating by the ability to lead and gain the assistance and cooperation of others in a team endeavour.

    Ability to work in a multi-cultural environment with sensitivity and respect for diversity.
